The movie follows the novel's investigation through its resolution, but cuts subplots and changes the final scene.

Crawford's dying wife was cut from the movie
The novel follows Jack Crawford's private life as his wife is dying. Screenwriter Ted Tally tried to keep that story through his early drafts, then removed it to keep the movie focused on Clarice.
The same decision reduced the background Harris gives Jame Gumb. The book can enter Crawford's, Lecter's and Gumb's thoughts. Tally wanted the movie to reveal much of the case as Clarice discovers it, so Gumb's past receives less explanation.

Lecter's written threat becomes the final scene
Near the end of the novel, Lecter writes to Chilton and threatens him. The movie turns that threat into action: Lecter follows Chilton through a crowd after telling Clarice he is having an old friend for dinner.
Demme said the book's closing letters would not work as a movie ending. He discussed the change with Harris before filming it.
